This video will show you how we collect your DNA for DNA Art using a buccal swab. First, we extract your DNA from the buccal swab and suspend the sample in a liquid buffer. This extracted DNA is then added to a PCR cocktail or "master mix". This master mix includes the primers specifically designed for your genetic theme you choose in the Ordering Process. It is then placed into a thermocycler for amplification. This is the Polymerase Chain Reaction (PCR) stage of the process, where the DNA goes through 35-40 cycles of denaturation, annealing, and elongation. During this process we load the DNA into an agarose gel and run a current through the DNA samples. Because DNA is polar, the fragments are attracted to the negative terminal of the machine. This step is to ensure the integrity of the DNA before we continue. While everyone's DNA is truly unique, there are certain areas of your DNA that you share with every other person on the planet. Since scientists know the order or sequence of those bases, they have developed chemicals known as restriction enzymes that seek out those spots. Restriction enzymes find those spots that everyone has and the enzyme cuts the DNA strand into two parts to get a unique DNA fingerprint. We now load the DNA a second time into an agarose gel and begin gel electrophoresis to obtain an image of the rate at which the fragments move and then analyze that information to determine the size of the fragments. This is the final step in analyzing DNA fingerprints.
