i had to do mine this week. i did mannitol and novobiocin for my pos and figured out it's S. aureus and for my neg I started with the SIM and then did urease so I know it's P. mirabilis.
At my school, most of the money is given to a&p, so b/c we use so much money on media, we don't get gloves. people brought it the first week but no one uses them anymore.
Oh and my 2nd week, I spilled mixed culture all over myself.
Gold star lol!!
Enjoyed your vid :) good luck with school

Always open the plate besides a blue fire!
You should have learned that, didn't you?

@danifilth3
As you were able to see I had already identified the culture of bacteria I was working with so aseptic technique wasn't necessary as my purpose to that colony had been done. As for your question in regards to "you have learned that, didn't you"? It is an immature and very unprofessional on your part since working aseptic technique under the flame has its purpose and none of them include opening a completed and disposable TSA plate.
Go study some more buddy ;)

on a tsa plate, the streak turned pink...but my gram stain turned out wrong...cuz its not showing it as a gram positive...its showing up gram negative.

@buttrpecan9 The GRAM Stain is the most important step of finding out the bacteria you're working with because you have to go either testing for GRAM + or GRAM -. You must get the the GRAM Stain right first and foremost. After you know what type of organism you're working with than you can take a step into doing the other tests and so on.
