IIRC this was an unbilled cameo, so Culp's appearence was probably a big surprise to the original viewers.

Was it because they would drink the same drinks on I Spy, or because the entire scene is a direct parody of their banter on the series. I never quite got it.

Nancy wasn't his sister. Robert was an only child.Nancy spelled her name Kulp, not Culp.Nancy passed away a few years ago.
