does it have to be Raw format in order to do this adjustment?

The adjustment brush works with any file. You'll have a lot more room for adjustments if you use the raw file, but the brush works on JPEG and other images, too.

Is it possible to make multiple adjustments on one selection? Whenever I try to, say, bump up the clarity and the exposure, It will only do one or the other. Is this just the way it is or is there something I can do to fix this?

The trick is that you have to "show the effects sliders." There is a little slider-switch to the right of the adjustment. Normally, only one adjustment is applied at a time and you choose the effect from the drop down menu. If you want to use multiple effects, click the effects slider to the right to activate it. Now, you'll see controlsfor all the possible adjustment effects (6 sliders and one color picker). You can adjust these sliders before or after you create your adjustment area.

I have the same setup (Vista and Firefox 3) and the sound works fine for me. Are you still having trouble with it?
