Type II topoisomerase is essentail for DNA replication and chromosome segregation. It relaxes supercoiled DNA by forming a covalent DNA-cleavage complex that allows DNA transport through a double-stranded DNA breaks. The intermediate complex was captured by the addition of antibacterial quinolones, two molecules of that intercalate at the highly bent DNA gate (3FOE.pdb; Ref. Laponogov et al. Nat Str Mol Biol v16, p667).
