Thank you so much for this! If a publisher does not want anything but the manuscript and a cover letter (no query) should the cover letter include a synopsis or summary?

Thanks for your help. I was recently asked to write a short story and send in a manuscript. The story is only suppose to have 150 words. Just to be sure, may I ask you, Should I write page 1. and this is page one. Page 2. and this is page two...ex... all on one or two 81/2x11 sheets of white paper?
