In the movie they look down on him because they think he is just a dumb homeless black guy and they treat him like he's an idiot. Eddie Murphy looks at the camera to show that he knows they think he's dumb and (like the audience) he doesn't like them either.

Breaking the fourth wall, as this is called, has been done for decades. It's been done in dozens of films, including The Wizard of Oz. Google "TV Tropes breaking the fourth wall", there's a massive list on the tvtropes wiki. (You will spend HOURS on that web site, I promise you.)
