@GherkiKnight After you escape the Labyrinth (Defeated the Island's core and can return to the Sand Zone) Jenka replaces the Life Potion if you've used it. She will do this as long as you're at a point that you can return to her (i.e. any point between escaping the labyrinth and before the final boss fight.)

It being brought to my attention that one of the wonderful soundtrack pieces (a Leitmotif, no less.) of the brilliant game Cave Story was based on a culturally significant piece of music, oh internet, the inter-connected-ness of things astounds me, even to this day.
