The movie is very Japanese and made for Japanese audience but the misery of peasants and superficial glory of samurai are the same elsewhere in the world. I think that's what makes this movie enjoyable to watch for many regardless where they come from.
Japanese farmers weren't that defenseless however. First ruler to unite Japan after the Sengoku era happened to be born out of peasant family and he knew the danger. Naturally, first thing he did was banning all the weapons from the farmers.

For me, the entire film is my favorite scene. You just can't go wrong picking any of the scenes.

This is my second favourite scene, the best scene I think is when the rice saved for the samurais is stolen, and the farmer who was supposed to be guarding the rice is picking up the leftover grains from the floor crying, and at that moment Kitsushiro throws down some money so he can buy some more, but tells him not to tell Kambei.....
My favorite scene was the part where they find the samurai armour and they samurai feel angry, and then shame
