LOST Season 1 Episode 3 - Tabula Rasa. A clip highlighting the end scene of the episode portraying John Locke with a sound similar to the sound of the Smoke Monster. Season 6 reveals how these two similarities link.
John Locke was not the Monster though, he was a host. I think this scene shows how much closer Locke is becoming to the island gradually, and how much further he is going from the survivors. If you look at season 3 and 4, Locke is still caring towards the survivors, but unlocking the mysteries is his main goal. After Walt leaves the island and Boone dies, Locke no longer really has "friends", and his relationship with Jack becomes increasingly hostile.
