I prefer to go full-out for the drivers, but it's up to you what method you prefer.
If you kill the Gang leader, you will automatically defeat the jeep without needing to destroy the vehicle or kill the gunner.
Please, get the capacity upgrade. I know the AK's just one Rep Level away, but since you're not there yet... The replay denies me that, so I can't help it, but you should have enough cash.

Unfortunately not. If you aim directly at someone Tony will refuse to shoot, and if you use explosive weapons or cars they will just get up afterwards unharmed.

@MaxAndEmilytate If you switch to his Henchmen, though, you CAN mercilessly kill random bystanders. It's fun to beat people with Pipes using his Driver then hear him say "What do I have to do? Bash your head with a pipe?" or something like that after killing somebody with a pipe.
