Dominic Toretto (Vin Diesel) and his new crew; Letty (Michelle Rodriguez), Tego (Tego Calderón), Rico (Don Omar), Cara (Mirtha Michelle), and Han Lue (Sung Kang), are hijacking fuel tankers in the Dominican Republic. Dominic begins to suspect the trail is too hot and leaves Letty to go elsewhere to avoid being caught. In Panama City, Dominic gets a call from his sister, Mia (Jordana Brewster), who tells him that Letty has been murdered after getting into a near fatal car accident. Dominic heads back to Los Angeles to examine Letty's crash and finds traces of nitromethane. He then goes to the only car mechanic that uses nitromethane and coerces him into giving him the name David Park, the man who ordered the fuel. Meanwhile, F.B.I. agent Brian O'Conner (Paul Walker) is trying to track down a drug dealer named Arturo Braga. His search leads him to David Park. Dominic arrives at Park's apartment first and hangs him out of the window by his ankles before letting go. Brian, who was also on his way to Park's place, saves Park and Park becomes the FBI's new informant. Park gets Brian into a street race through Los Angeles; the winner will become the last driver on a team that traffics heroin between the United States-Mexico border. Brian selects a modified Nissan Skyline GT-R R34 from the Impound Lot, and Dominic races with his modified 1970 Chevrolet Chevelle. Dominic shows up to race as well, during which two drivers are killed. Dominic wins by bumping Brian's car, making him lose control. Brian uses his power as an FBI agent to arrest another driver, Dwight Mueller, and takes his place on the team. The following day, the team meets Fenix Rise (Laz Alonso), one of Braga's men. It was revealed to Dominic that Fenix was the person who murdered Letty. They drive across the border using underground tunnels to avoid detection. Brian had prior knowledge that after the heroin was delivered, Braga ordered the drivers to be killed (he was able to plant Letty undercover to capture Braga in exchange for Dominic's freedom when she was murdered). After a tense stand-off, Dom detonates his car with nitrous to distract Braga's men and Brian hijacks a Hummer with US$60 million worth of heroin. Both Dom and Brian drive back to Los Angeles and hide the heroin in a police impound lot where Brian picks up a modified Subaru Impreza WRX STI. Brian tells his superiors that in exchange for Dominic's pardon, he will lure Braga into a trap, forcing him to personally show up to exchange money for the heroin. At the drop site, however, the man who claims to be Braga is a decoy and Campos (John Ortiz), the real Braga, escapes and flees to Mexico. Brian and Dominic travel to Mexico on their own to catch Braga. They find him at a church and apprehend him. As Braga's henchmen come down to rescue their leader, Brian and Dominic drive through the underground tunnels back to the United States. When some of Braga's men are killed, Brian crashes his car and is injured after being T-boned by Fenix at the end of the tunnel. Before Fenix can kill Brian, Dominic, who survived the explosion and exchanged his 1970 Dodge Charger with a 1973 Chevrolet Camaro, drives into and kills Fenix. As police and helicopters start streaming to the crash site on the U.S. side, Brian tells Dominic to leave, but Dominic refuses, saying he's tired of running. Despite Brian's request for clemency, the judge sentences Dominic to 25 years to life. Dominic boards a prison bus that will take him to Lompoc penitentiary and as the bus drives down the road, Mia and Brian, along with Tego and Rico (who assisted in the Dominican Republic heists), arrive in their cars to intercept it. (An animated sequence is played during the credits, in which Brian brakes in front of the bus causing it to flip)

Dominic Toretto (Vin Diesel) drives the streets of L. A. as if he owns them. As far as his crew is concerned, he does. He spends his days putting wrench-time into high performance racing cars - make and model matter less than the computer-controlled fuel injection that makes them fly. Nights, Dom pilots his own earth-bound rocket, pocketing as much as $10,000 a ride when somebody has the nerve to race him.
The races are street theater, tribal gathering and battlefield, fuelled by adrenaline, sexual tension and raw, out-of-control speed. Throngs pack otherwise desolate streets to cheer and to jeer and to belong. Dominic struts through this high-octane outlaw scene like a rock star, presiding over the hungry roar of engines, all set to test the limits.
Brian (Paul Walker) is hooked, hungry and ready to test the limits, too. He prides himself on street smarts, but to this crowd he's white bread. He seeks Dominic's approval behind the wheel of his own NOS-injected muscle machine. After a blazing encounter with ruthless Johnny Tran (Rick Yune), Dominic decides Brian is all right. Dominic's sister Mia (Jordana Brewster) likes what she sees, too. The thing is, they don't know he's a cop.
Brian is under deep cover, investigating a series of sensational big-rig hijackings. The police and FBI need it to stop before the truckers take matters into their own hands. Cash flows freely through the street-racing scene and the cops know some of that money is dirty. Dominic and Johnny are both suspects.
As the rivalry between Dominic's and Johnny Tran's crews escalates to ever more dangerous levels, Brian's bonds with Dom and Mia deepen. He feels a genuine kinship with Dominic, and the attraction between him and Mia is too strong to deny. But he's still a cop and as the pressure to wrap up the hijacking case intensifies, Brian must decide where his loyalties lie and what his limits really are.
