In the canon, Holmes is 27 years old when he and Watson meet, and Watson is a few years older (all of this is deductions made by afficionados of Holmes and can be "verified" in any reference book pertaining to the canon).

One change I need to mention is the rape scene seen in the 10th section of this movie. This does not occur in the book. There is no inkling of such an occurrence and it's too bad that this was unnecessarily added to an otherwise excellent adaptation of one of Dame Agatha's stories.
