I think he knows it's not really his sister but his girlfriend "being" his sister for Halloween, and the kiss is just meant to creep out or amuse the viewers. Of course, in real life the actor kisses Jennifer Aniston, not Ami Dolenz or whoever the girl "underneath" is played by, but he's an actor and they're both actresses and it doesn't really matter unless he was dating or doing one of them...
