Each key on the keyboard/piano or each fret on a guitar/bass that you move up or down is a 'semitone' or 'half-step' up or down. He means that the tuning is one half-step flatter than standard tuning. Drop your 6th string down until the 4th fret matches the tone of the open 5th string, then tune the rest of them normally to that.
