Did all that was required and the tyre is still not inflating. Is one cup of sealant enough. Should I pull the whole lot off and start again.

I think you are mixing up "Tubeless" and "Tubular".

One that looks like tireless is called "Tubular tire"

"Tubeless" looks pretty much what the name says, "Clincher tire without using a tube"

Additional advice guys, to put the sealant in you can actually infect it through the valve. Most sealant companies include a sealant injection. Further more, do not use a tool to put the tyre on the rim as it can tare it or chip away bits of rubber, just use your hands. This is info i got from other youtube vids.
