If you look at the facts overall I would have to say river would win. Yes Buffy is durable strong fast and has good reflexes but river is a million times smarter and she's PSYCHIC not only can she tell what Buffy is thinking she can see her every move. Eventually Buffy would make mistakes and river being the super genius she is would find a way to kill Buffy instantly. Even the slayer can't recover from death.
