Drupal: Introduction to Panels

How can I theme panels? Adding a different highlight instead of the same one that all the other blocks have? Any idea? Please help, i tried css id and everything there must be something I am doing wrong!

Thank you for this helpful video. I have a problem, however. I get the following message when I try to add anything to a panel using panel 3 and drupal 6:

An error occurred while attempting to process /QA/panels/ajax/add-pane/1/top

Can you help me resolve this?
