DOS = Denial-of-service and it only 1 guy that is attacking a server
DDOS = Distributed denial-of-service and with this alot of people are attacking atleast 100 and those people who are attacking are infected and don't know they are actually attacking a server
According to me, it is very difficult to detect a source of DDOS attack considering the present resources. Caroline pointed out a very important fact which is worth looking into, as TWITTER's distribution system is pretty fragile as compared to to others like Facebook. This can be due to cheap or low number of caching servers used by Twitter (they have had no business model for years !) i.e. lower built-in redundancy.
