As for Raiden, as someone put it, his "epicness" came from the aftermath of MGS 2, and in between MGS 3 and 4, he seeks the help and guidance of Matka Pluku, AKA Big Mama, and Snake's surrogate mother. He endured training from her in order to prepare himself for the "new age of technology", as his former Colonel Campbell described it at the end of MGS 2. The origins of his artificial blood and mostly cybernetic body is anyone's guess, but I do believe it was his choice (don't quote me on that)

Okay, it's not that Vamp is a legit vampire. That would be ridiculous, even for an MGS game. The truth is, Vamp has this wacky "condition". His nanomachines were modified from the original version. I don;t know the details surrounding this fact, but Naomi altered his nanomachines to make him superhuman at the behest of his request. But the nanomachine modification was, as she put it, "unstable", because she couldn't find the correct way to supress them once they spread throughout his body.
