@DarkSlayerRiku Beat story mode with 3 turtles i.e defeat Shredder with 3 turtles first. Then with the 4th turtle, get to the final chapter and defeat Shredder. You are then taken to this stage. ;)

Its locked. You need to unlock it. It will be unlocked once you've completed story mode with all 4 turtles.

Oroku Saki is pretty tough..Even if you have 6 lives :( but Gembu Mode saved your Shell :)
