The reason he is making that noise is because he does not like the noise you are making, it bothers him/her so it makes that grumbling noise in distaste

If it's a low sounding purr and they seem in a relaxed posture its just thats its comfortable. If it was unhappy/displeasured they would make a higher purr sound and it feel like the guinea pig is vibrating.

I didn't know that was purring. I had many guinea pigs for many years as a teen and to me it was always just a sound they made when they were horny. Although sometimes the girls would do it to each other and now that I know it was just purring I feel a whole lot better about that. (I was really homophobic back then).
funny, sometimes my guinea pigs purr like that when i give them their fave food. I dunno, all piggies are different. Maybe this one its a sign that its happy!
