I think Twilight squees in Feeling Pinkie Keen when she jumps away from the Hydra and slams into the side of the cliff... :D

Why does the flute pinky pie takes have holes on top? HOW would they use the flute in such a manner that the holes still make sense? Is there a spell to give them fingers?

It sounds like Bubbles from the Powerpuff Girls to me. But the voice actress who plays Twilight was Bubbles, not the one who plays Fluttershy.
Unless you're referring to the noise she makes when she takes off running; that sound effect is used all over the place.
