No Sypha is a guy. In the japanese version The ending still refers to Sypha as a man. HE and Trevor remain "close". Trevor was always the fruity one in the family... but still my favorite Belmont. They even make reference to it on Castlevania Curse of Darkness while he fights Isaac, in Japanese of course.

Early on it was intended Samus was a guy, but the cold hard truth is revealed after you finish the game in the earliest possible time, or by using the infamous Justin Bailey code. There it is revealed we see Samus's true identity, a female, dressed in a purple leotard and has long green hair.

PS: Next to Trevor and Alucard, Sypha is one of my favorite characters in that game. I rarely use her but she IS a useful character to have join you.
