Awesome! I'm having trouble sectioning brain tissue in paraffin. I always end up with wrinkles or some kind of puckering when I mount on slides and also the sections roll over on themselves while I'm slicing. I read you suggesting refrigerating the blocks beforehand? help please!
Thanks, All I can say about that is practise and practise. My first coupes where really bad but after a lot of practise it became better and better. Important is to cool the block in the fridge.

@RONALDSCHULTE Ik heb nog helemaal geen ervaring met snijden. 1 keer geoefend. Er moet nu veel gesneden worden dus iedereen wordt een dag ingezet net zo lang tot alles gedaan is.
Also, my tissues seem to be shredding apart when I cut. How can I fix that? Is there a website I can go to to get some tips on cutting, mounting, and staining?
Nice clean ribbon! Why can't my microtome make something like that? I end up wearing most of my ribbons or it ends up all over everything(including the microtome) XD Cooling the block doesn't help nor does changing the blade. I guess it's the kind of paraffin wax we use or something.
I use paraplast but use of normal paraffin is fine to. Of course is my cutting not always a party. Speed, umidity, temperature, thickness etcetera. Al those things must be perfect. When the air is to dry you can't cut one good coupe. Lung tissue gives better results than a hard Sternum. Start with 7 or 8um. That should always give a good coupe. If not than is your knife angle maybe to great or small. My angle is 32 degrees. Gr Ronald
We have been just using normal paraffin so far. I try to keep my ribbons 5 micrometers thick and I try to average one slice per second. Lung tissue is pretty easy, and I found kidney tissue wasn't that bad either(but that could just be me or something). I make sure my clearance angle is around 2.5, it doesn't go higher than ten or something. I know it's not the actual degrees. We do everything in room temperature but we keep our blocks on ice when we're not using them.
I always using the needle to spread the cutting tissue , but I will change to the brush because I always cutting the tissue with the needle

Not sure if you were cutting slowly for demonstration purposes but I would recommend cutting faster to get a better ribbon.
