Both guitars play parts that are too high to play on a bass. Also, around 1:53 you can hear a bass playing and neither player has notes.
To answer the original question, it looks like they switch off leads, but I bet the game calls the right guitar the lead part since it's less repetitive overall.
Actually the funny thing is that the one on the right is rhythm, and the one on the left is lead. No joke, my friend and I played it and he's pretty shitty, so I said "okay, I'll play lead if you're gonna puss out on me." Needless to say, he was crying halfway through the song.
