I have read the work of Paul Ekman and I myself have seen the very same body language they describe when they say someone is lying.... This work is based on very firm foundation. Although it's not always easy to infer what some people may be hiding or why.

Anyone can pass a polygraph as long as the probable-lie questions, such as something people would normally lie about (have you ever stolen money) have a higher "spike" than the relevant questions.
By just math, you can calculate how four spies can breach security in a government agency. Let's say you screen 10,000 employees, with a 90% polygraph accuracy. 10 are spies. You catch 9 spies, but about 1000 people lose their jobs for a false negative. You screen 1000 replacements, 10 spies 1 gets through, 100 honest people are denied employment. You screen 100, 1 gets through, 10 don't get hired. One gets through, 1 honest person is hired. 4 spies are hired 1110. It's less than 90%.
The best part about this intro was with the "gestural retreat" where he backs up, crosses his hands and that's supposed to be a sign that you don't believe what you are sayin... and they show the "I am not a crook" Nixon speech where he does the same thing XD XD

Some very huge flaws in Polygraphs. I personally known someone who took the Poly test for her application to a local police department. She was asked about sexual relations with a minor and about incest. She failed miserably on these two questions. She was the victim of sexual abuse by her step father when she was 9...it doesn't take a genious to figure out that these questions are going to provoke strong physiological and emotional response.
There are also those who are well aware of the polygraph flaws - it is unsettling that some machine may falsly indicate you are lying - The fear of this alone can create false positives. Then there are those who have social anxiety disorder...simply being in a room one on one with a person is stressfull

The question then becomes, how do soldiers in the field know who to stop and who to let go? In reality, it's tough to figure out who's a bad guy and who is not in that particular environment. Bust everybody in the village? Unrealistic. Same as using a TV show to prove that the instrument used is BS 100% of the time. You want terrorists caught or not? You sometimes break a few good eggs to weed out the bad ones. If these people didn't want us coming there, stop the bad guys from living with you.
That doesn't mean for a second that we should stop looking for and testing new means of achieving the desired result with the smallest loss ratio possible.
If we didn't, then we'd still be using sticks, rocks, and ... tasers. Oh shi-

It could work in the same way a copy machine lie detector work.... Point at suspect, they believe it works, they give useful information. The operator would have to use it that way, though.
Too bad federal, state and local law enforcement agencies use the polygraph (and other failed "lie detectors like voice analysis) to screen applicants. The public at large is learning about the pseudoscience behind polygraphy at an increasing rate and this utility is disappearing quickly.

No. There is no "Pinnochio reaction" that can be read on people's faces when they lie. However, facial expressions may give clues that a person's emotions don't match their words, and in some such cases, inferences might be made about whether a person is telling the truth.
I think if you watch a brain scan of the person while asking questions, and if certain parts of the brain light up (like the ones that involve imagination), you can tell. But if you convince yourself that you're telling the truth, I'm not so sure that would be a good way to tell either.
There is a Mythbusters where they test different polygraphy methods. One of the tests they did was the brainscan test. Grant actually managed to lie through that test by thinking happy thoughts.
Just look at him, he is so stressful, he is not confident during all his time of talking. Why is it so??
