They knew it was suicide for two reasons. First the tape was already played back for them to hear it. The second reason has to do with the fact that they heard the gunshot on the tape. If they heard it, meaning he was dead, he wouldn't be able to stop the recording. It would still be recording which it was not.

The answer to the riddle is that the tape was already rewind to the location of what the police heard. If it was suicide the tape would have recorded initial it ran out of tape and then stopped. Since it was at the location of the message some key had to rewind the tape to that location. The dead person could not have do it.
