"Yasu is the culprit," comes from a very old video game in Japan, in which you play a detective trying to solve a series of murders with your partner, Yasu. The murderer does turn out to be Yasu himself, making it one of the most familiar and old plot twists there is. Kyon saying "The culprit is Yasu," while Asakura talks is basically him saying that her 'plot' is so cliched he can guess what's going to happen.
