wow, was just reading about viewing cytokinesis with GFP and chanced youtube for a vid - thanks for the upload, much appreciated. What kind of microscope, software and hardware did you use to view and record this?
I used a TE2000-S eclipse Nikon microscope, which is a fancy microscope often used for fluorescence imaging. It has components built into it so I can shine specific wavelengths of light on the cells. A QImaging (QICAM 20775) camera is attached to the microscope so I can view the cells on a computer screen. Micro-manager software works together with the camera to take pictures of the cells. I then take a picture every 15 seconds and use ImageJ software to make the movie.
