  • Furthermore this video should emphasize the use of dATP-αS instead of dATP for elongation to prevent the nucleotide from being degraded by the luciferase prior to being added to the nascent strand.

  • The video has an error the Apyrase takes up the dNTPs faster than the polymerase attaches them so it should illustrate that. Also Pyrosequencing does not work well with multiple repeats such as AAAAAA because the Apyrase gets rid of the dNTPs too fast
