@vortexlisa you can get that door opened if you don't kill the Robot Engineer in the mission just before. That engineer is the civillian you get almost at finish. Just leave him unconcious and he'll open the door on next mission!

To open the door at the end of the hallway, the technician by the Interceptor in Area 51: Infilitration must remain alive. This means knocking him out. Afterwards, center the aiming reticule in the center of the door where the crack is. This should cause said technician to open up said door kindly asking you to leave said area.

The difficulty does not matter - as long as he is knocked out on Agent (or any others), he will appear for all 3 difficulties.

The guy from the previous level that gives you the lift key card unlocks it for you. You can't kill him on the previous level though, so knock him out to get his keycard.

This level was a downright pain. I cannot tell you how many times I got killed by the blue uniformed guys. Trouble is, you might kill all of them while t=you are fighting through those areas - but there are one or two blue guards that constantly respawn. So on Perfect Agent, if one of them runs up behind you and you get Dragon'd, it's pretty much over.
