To be specific, the difference comes from the gin. The aromatic components are dulled if the drink is cooled below about thirty degrees. If you shake the drink, the gin gets chilled.
However, it should be noted that this is only a concern with GIN. With Bond's vodka martinis, shaking the drink has no detrimental effect, since vodka has no components to dull. Therefore, all that happens is you get a cold drink.
In order to get the proper effect, they'd have to use potato vodka like the stuff from Fleming's era. Shaken vodka tastes less oily because of the dispersal of the oil during the shaking. It also makes the drink weaker.

Look at the olives, the floating ones are In the shaken one, the sunken ones are in the stirred.
The shaken olives float because there is more water, making the drink less dense (as alcohol is less dense than water)
