This is not "Pokey"'s theme... This is Giygas's theme. One look at its japanese title (Inakunarinasai) will tell you that. It was named after a line that Giegue/Giygas used in MOTHER. In which he told Ninten and his group to "Dissappear" or "Cease to Exist".

Giygas is the final boss of Earthbound, a formless, swirling mass of pure evil who gives people NIGHTMARES.

Most of Earthbound from what I hear is crazy and kiddie (there's a prophetic apple in there somewhere), the nightmare fuel that is Giygas is very incongruous, which only serves to intensify its scary effect.

If you're gonna make such a big deal out of it you should know that Pokey Means Business is not the real name of the song. That was just something made up by SNESmusic when they ripped the game audio. If I wanted to use the official name I would call it イナクナリナサイ (Inakunarinasai), but I'm not going to do that, because about 99% of people who've played Earthbound just know it as the song that plays when you fight Giygas.

Oh god that would be impossible. The Guitar Hero Music Studio interface is pretty terrible. It takes about 5 hours to do what takes about 15 minutes with Garageband and FeedBack. Plus it doesn't let you do tempo changes, which this song uses a lot of. And you only have about two octaves worth of notes at your disposal and all the instruments sound like they're from an 80s adventure game by Sierra. I did do a cover of Pollyana from Earthbound Zero, though. You can review that if it's still up.
