It's a difference in focus imo. Ledger's Joker is a comedian, he's trying to make a point, some sort of grand commentary on human nature; when he laughs, he's laughing at the absurdity of the world. Hamill's Joker (and many other interpretations) is a clown, he's looking for something more primal and basic, and any opportunity for a joke is one he'll take, even to the degree of swinging wildly between harmless, petty crimes, and mass murder.
