As they multiply, the rate of multiplication reduces because of the using up of the nutrients in the agar, right? Is that why as time passes, the bacteria colonies seem to become lighter and lighter in colour?

No Fong, the agar was drying out, as well as the bacteria.

this was my very first attempt at making movies of bacterial colonies. I was trying to find out the conditions for making such movies. Drying out is still a major problem. My latest movies are better but havent posted them yet.

why would anyone want to watch a plate dry out over time... it'd b much cooler if you did one of the colonies growing
