Shino was laughing because he was testing some food to make sure it wasn't poisened (the bugs would have broken down the poison). It wasn't poisoned. But there was a laughing potion in it. Therefore, Shino bursts out with laughter for, as far as I'm aware, the only time in the entire series. One of my fave episodes btw. :)
