Most likely he was watching when the guy looked at his cards. Some players may raise their eyebrows, sit up straight and/or look at their chips when they pick up a good hand.

The first hand was pretty easy to read, since both paired the aces. Only thing negreanu had to worry about is if someone had a mad hand with AA or A plus a paired card on the flop so it would improve their chances of winning on the turn or river. Either way you get the big advantage percentage wise, and any other hand other than pocket 5's is a loser with those given percentage odds.
