Hey just wondering, I am doing this in my next Extended Experimental Inquiry- because Congo red is carcinogenic- would using blue litmus dye for the yeast work? Obviously the colours would be reversed (with blue litmus dye... blue = basic, red = acidic). Thanks

@Shydrasnake The title that come to my mind is called "How to know the protozoa" H.E. Jaques. I was using a really old edition dated 1949. They probably have newer editions by now but that was the title of the guide I used to learn most of what I know at this point.
@chaosarcee Still water works best. Try a pond or a really old puddle. If it's stinky, that'll tip you off to the fact that "somebody is passing gas" & you should find someone in there. Good luck.
i got some Chaos (Pelomyxa) carolinensis from Carolina Bio supply--- i've seen paramecium in the culture, but there are these other long things about the size of paramecium, that sometimes suck up food, and sometimes open their heads into too circular 'weapons' with super fast rotating cilia (like chainsaws) that whip food into their mouth----
1) I made sure i had a happy bunch of paramecium.
2) I put 1/2 envelope (1tsp or so) of regular bread yeast in 100ml warm water. Then I let it sit somewhere near 37C for a couple hours, till I saw a lot of bubbles in the center of the glass.
3) I added about a dozen or so drops of 1% congo red dye into the glass.
4)Meanwhile, I took a sampling of paramecium onto a depression slide. I waited a couple hours before I applied the congo red yeast.
Is there any way to "pick up" and move these microorganisms? I've tried to do it several times and ended up just moving them a bit to the side or squashing them.
I use a disposable plastic pipette. You could try one of those "booger bulbs" folks use on babies to suck up your protists and put a drop or two on your slide.

The original culture was P.caudatum. By the time I had shot this I had subcultured them several times. They still look like the P.caudatum, but evolutionary processes being what they are, who knows what they could be turning into
This type of dog is the Devilspjhi dog. This is young and cocky with a small organ. Mostly over eats therefore with poor interacting skills. This type will stay this way and if it finds a mate will be of like nature.
Ok....so i notice that after the yeast is directed into the oral grove by the cilia and forms into food vacuoles at the gullet they turn black as they get futher away from the gullet....so thats probably the yeast being obsorbed/digested.....are the black things just shells of a food vacuole that get excreted or is it still yeast?? what do you think?
Sounds like you have the process down. The pH shift and the color change indicates digestion in progress. Eventually, the wasted remainder does get excreted. I didn't get that on video, but if you do a search you will likely find someone who did.

well, some micro-organisms actually have mouths! But, to capture their "prey" they can use a bunch of ways to get it. although, if i'm not mistaken, paramecium have cilia(hairlike structures) to suck their "prey" into the mouth. and if it's a solid, then the "prey" goes to the food vacuole. and that's that!
The paramecium have what is called an "oral groove". It's the mouth area that takes in whatever they choose to ingest. Unfortunately, the oral groove isn't very visible on this. If you look at other images of paramecium you will see something that looks like a slit or valley from the center of the animal to the anterior region. that slit or valley is the oral groove.
