My general rule of thumb for pick strokes is pick down on the downbeat and up on the upbeat. For Spirit in the Sky, my hand is always moving in a steady eighth note rhythm (1 and 2 and 3 and 4 and...). So, my hand is picking down on the numbers (downbeats) and up on each "and" (upbeats). Hope this helps! Sometimes, its best not to think too much about these things and just go with what feels right.
