Yes. This film is based loosely on Homers' Odyssey. On the opening scene the Cohen brothers make sure to credit Homers Odyssey. The point of this edited clip is to show how the train hopping scene, car chase scene, robbery scene, shackled prisoners, and finally the title of the book, were obviously inspired by Sullivans' Travels. These scenes even occur in the same order in both films.
The Main similarity is the name. The title of the latter movie came from the earlier one (the name of the movie they had been working on in Sullivan's Travels).

I believe theCohen Brothers based Oh Brother on Sullivans Travels. Both great films, but a totally different theme.
