A professional level of your craft is usually assumed and doesn' t have to be mentioned especially - visiting a dentist you won't ask if he masters his craft.
But as pointed out above, its not so much how good and skilled U are (as thousands of people show an extraordinary level of artistic mastery). Needless to say, everybody in this field is willing to kill himself to succeed...

Question: Is this painted with multiply setting on the brush tool? What kind of settings allow this sort of smooth flow with a chalk like highlight?

It depends mostly on 2 things to succeed:
1. Who U know
2. Being at the right place at the right time

Sketchbook Pro, made by Autodesk/Alias. It simulates drawing very well. You can draw this type of stuff with Photoshop or Painter as well.
