You have several options for this sort of thing. Directly modding a Genesis ROM would probably be the hardest due to all the obscure file formats that would likely be involved; I'd recommend building the game in SORRMaker (once that's completed, it's supposed to debut with SORRemake version 5). In the meantime, there's also the Beats of Rage game engine.

is there any possible way to make a streets of rage 2 game but put characters you want in the game like ryu or kyo or anyone you want?
