On the HEK plate, most gram negatives will grow in yellow/orange colonies. Salmonella will grow in clear colonies with black spots on the top (they produce H2S) and Shigella will grow in green colonies. This media is mainly used for stool cultures. There are a few different sugars in this media, the exact mixture, I am unsure of.
MAC is used for the differentiation between gram positive and gram negative. Gram negative bacilli to be more specific. Beyond that, it only selects between lactose fermenters and non-lactose fermenters. Which, is sometimes helpful, but not usually. There is MAC with sorbitol (replaces the lactose) also used for stool cultures and differentiates between E. coli O157H7 and regular E. coli. Regular E. coli's ferment sorbitol and O157H7 does not.
Love the bacteria song. Was a bit horrified that some people were holding test tubes and plating bacteria and not wearing gloves some people wore gloves but not everyone. So Dangerous!!!!!!

Some people are more likely to be careful about washing their hands and what they touch when they aren't wearing gloves instead of being careless because they think it doesn't matter if they're wearing gloves. Usually when you plate a petri dish, you don't really touch anything with your hands. The flamed loop is supposed to be sterile. Without gloves it is definitely not 'sterile technique', but you could think of it this way, the bacteria that would be on your hands from touching surfaces would also be on the gloves.
Therefore, it's really to protect yourself. In microbiology lab classes, I've used some pretty nasty bugs and definitely would not want to contaminate my books, bags, and food with them.
Also something interesting to note, the red petri dish that has the the clear smear, is probably bolvine blood, and the bacteria that caused the clearing uses the nutrients in the blood as a food source. Probably not a good thing to have on your hands.
Yeah, true. But I'm sure a classroom environment, such as seen in the video, aren't handling anything more than a BSL-1. More protection is needed as you go up the BSL.

the black media looks like BCYE (buffered charcoal yeast extract) and the green is most likely Hektoen (for the recovery of salmonella and shigella)

you don't need gloves for anything you do with micro. Just make sure you wash your hands after!

Not really, some bacterias need only few minutes to infect your skin cells than, when they are unwashable they get deeper and deeper.
