no, unfort, the ac2 savegames are locked. i have to back up the whole ps3 hard drive to replay most missions.
for the bonfire dlc, the assassinations can be done in any order, so its a little more convenient. i quit the memory right after the assassination so that i can replay. except for last rites - you have to quit the game during the memory corridor dialog in order to replay that memory.
its no big deal, but my guess is that ppl are pointing out that the target indeed dies. im not sure what you mean when you say the target is still alive cuz of the arrow. would you like to clarify?
oh no sorry i didnt see it right lol its just when u assassinated the target i was looking at the guy behind him at 2:35 cos he was standin there and i thought that was him lol soz my mistake :)

If itt had a replay feature, I would try to do my missions like you do...
Kinda like in the first game...

its a kill animation that occurs when the target has just engaged in fighting with you, but hasnt had time to put up his defenses yet.
