i think that you play as tommy from vice city. Also on the back of the bus van it says los santos. so that is the location, San Andreas :) in the docks it also says LS PORT - meaning Los Santos port? the beggining with the joggers there is a poster for a pier park which may be the pier where the ferris wheel is on GTS San Andreas? and the gym is where there is a beach gym in San Andreas :)

It's definitely in Los Santos but I think San Fierro and Las Venturas are gone. The other secluded area like the desert and mountains seem to be back. I think Rockstar created a bigger Los Santos and more secluded desert and mountain terrain.

11:25-11:34 the wall at the backgroung has big white letters. "LOS SANTOS", and we can clearly see that. It's san Andreas (or at least improved part of it). About the appearance of Claude, Tommy, etc. I dont think so. Rockstar said that they will not use the characters from GTA3 series again. There might be some references though.
