OK, so this is how the episode involving this song will go:

Everyone in Equestria will be put under a spell cast by a weakened Discord. The spell makes everyone into jerks, much like Fluttershy did. In order to revert the spell, the Mane Six, who are protected due to already being put under and breaking out of Discords spell, need smiles to defeat Discord. So, after many tries, Pinkie sings this song and creates a giant blast of happiness that destroys Discord for good.
