In so far as why Rachel uses the lighter in the manner she does I'm guessing that everything about her, like the rest of movie, is highly stylized (watch how Rachel carries her hands). It makes a jarring contrast to things that are familiar (the whole 1940s/Sam Spade atmosphere) to that which is unfamiliar. For me, this is the best Sci Fi movie ever made (though I love Aliens II as well. Who doesn't?)

This movie is a masterpiece: the synopsis, the plot, the script - and look at the way it's shot, the use of lighting - the light and dark and shadows during the entire scene, and the camera angles. It's so brilliant and original, I love it!

@route1box43 She needs to focus at the camera doing the test. She tries to light the cigarette and perceives that the lighter is off. She reacts to it with disdain at the same time she pays attention to that detail. It is ambiguous, her trend.
That is why everybody gets fascinated by this simple gesture in this scene.

@Micali711 There really isn't much difference between Replicants and proper Humans aside from emotions and lifespans and in the case of Rachel, at least one, if not both (depending on which cut you go by) , doesn't apply.
