We Use Bio-Rad Quality Control materials in the Chemistry laboratory i work for, and i have to say Bio-Rad has the best products and great customer service.

Check the linear range. Maybe your calibration curve is not good. Besides, Coomassie blue binds unspecific to proteins. So you won´t get a very accurate result.

Awesome video, Im a Biochemistry student, and watched this today between labs, made my day....Even though us scientists (well, Im an aspiring scientist, i guess) may have a stodgy image, we are just as messed up as everyone else, with a wicked sense of humor (when you do/learn this kind of stuff till ur eyes bleed, this video just becomes that much more enjoyable!)

Coomassie blue isn't really suppossed to measure protein concentration. Do a bradford assay. Then, make a calibration curve of just the protein and figure out the extinction coefficient.
