I admit the references are a bit vague at first glance, but I believe that is purposefully so. In my day (the 60's), it was absolutely scandelous to be other than totally heterosexual. As the movie is set in that period, it makes sense that references to Prudence's persuation would be left questionable.

Yes, she is Lesbian. Prudence is singing longing to her cheerleader teammate, the blonde one. Also, the musical call of the New York flat renters to their love-struck roomate Prudence, weeping in the CLOSET, is "COME OUT to play"; humorously poignant as well as deliciously tongue-in-cheek. Add to this, when they all meet up again at the Circus, Prudence happily remarks that her new ladyfriend is a Contortionist.
